KOH Preparation for Fungal Elements (Wet Mount)
The KOH Wet Mount is a medical chemistry test that detects fungal elements in a given sample. The samples can be skin, nails, hair, ear discharge, sputum, etc. This is a presumptive diagnosis of fungal diseases like ringworm, aspergillosis, dermatomycosis, mucormycosis, otomycosis, and so on.

- The specimen is emulsified in a drop of 20% KOH on a glass slide. The sample should not be bigger than 5 mm long and 2 mm wide.
- The slide is then heat-sealed over a bunsen burner.
- The smear is covered with a coverslip and left for 5-10 minutes.
- As soon as the specimen has cleared, it is examined under a clinical microscope. The fungal elements are demonstrated as shining elements in the smear.
